Light-induced dimerisation is the most commonly-used method in non-neuronal optogenetics. When exposed to light of a specific wavelength, a photosensitive protein domain undergoes a conformational change which then causes it to bind to an interaction partner to form dimers or oligomers. This video introduces three prominent dimerisation systems: the cryptochrome, the phytochrome and LOV domain-based iLID systems. TOPIC : AGROBACTERIUM MEDIATED GENE TRANSFER For CSIR NET/GATE/M.Sc./B.Sc. A. tumefaciens is a rod-shaped, Gram-negative soil bacterium which infect dicot plants. Some common species of Agrobacterium are- A. tumefaciens and A. rhizogenes.It is consider as natural genetic engineer because they transform the plant. To be virulent, the bacterium must contain a tumour-inducing plasmid (Ti plasmid or pTi), which contains the T-DNA and all the genes necessary to transfer it to the plant cell. Phototropins are photoreceptor proteins (more specifically, flavoproteins) that mediate phototropism responses in higher plants. Along with cryptochromes and phytochromes they allow plants to respond and alter their growth in response to the light environment. Phototropins may also be important for the opening of stomata and the movement of chloroplasts. Phototropins are part of the phototropic sensory system in plants that causes various environmental responses in plants. Phototropins specifically will cause stems to bend towards light and stomata to open. Phototropins have been shown to impact the movement of chloroplast inside the cell. In addition phototropins mediate the first changes in stem elongation in blue light prior to cryptochrome activation. Phototropin is also required for blue light mediated transcript destabilization of specific mRNAs in the cell.
