Fluid in the filtrate entering the proximal convoluted tubule is reabsorbed into the peritubular capillaries, including approximately two-thirds of the filtered salt and water and all filtered organic solutes (primarily glucose and amino acids) The loop of Henle is a U-shaped tube that extends from the proximal tubule. It consists of a descending limb and an ascending limb. It begins in the cortex, receiving filtrate from the proximal convoluted tubule, extends into the medulla as the descending limb, and then returns to the cortex as the ascending limb to empty into the distal convoluted tubule. The primary role of the loop of Henle is to concentrate the salt in the interstitium, the tissue surrounding the loop. Considerable differences aid in distinguishing the descending and ascending limbs of the loop of Henle. The descending limb is permeable to water and noticeably less permeable to salt, and thus only indirectly contributes to the concentration of the interstitium. As the filtrate descends deeper into the hypertonic interstitium of the renal medulla, water flows freely out of the descending limb by osmosis until the tonicity of the filtrate and interstitium equilibrate. The hypertonicity of the medulla (and therefore concentration of urine) is determined in part by the size of the loop of Henle. Unlike the descending limb, the thin ascending limb is impermeable to water, a critical feature of the countercurrent exchange mechanism employed by the loop. The ascending limb actively pumps sodium out of the filtrate, generating the hypertonic interstitium that drives countercurrent exchange. In passing through the ascending limb, the filtrate grows hypotonic since it has lost much of its sodium content. This hypotonic filtrate is passed to the distal convoluted tubule in the renal cortex. The distal convoluted tubule has a different structure and function to that of the proximal convoluted tubule. Cells lining the tubule have numerous mitochondria to produce enough energy (ATP) for active transport to take place. Much of the ion transport taking place in the distal convoluted tubule is regulated by the endocrine system. In the presence of parathyroid hormone, the distal convoluted tubule reabsorbs more calcium and secretes more phosphate. When aldosterone is present, more sodium is reabsorbed and more potassium secreted. Atrial natriuretic peptide causes the distal convoluted tubule to secrete more sodium. In addition, the tubule also secretes hydronium and ammonium cations to regulate pH. Though the collecting duct is normally impermeable to water, it becomes permeable in the presence of antidiuretic hormone (ADH). ADH affects the function of aquaporins, resulting in the reabsorption of water molecules as it passes through the collecting duct. Aquaporins are membrane proteins that selectively conduct water molecules while preventing the passage of ions and other solutes. As much as three-quarters of the water from urine can be reabsorbed as it leaves the collecting duct by osmosis. Thus the levels of ADH determine whether urine will be concentrated or diluted. An increase in ADH is an indication of dehydration, while water sufficiency results in a decrease in ADH allowing for diluted urine.

Nitrogen Balance = Nitrogen intake - Nitrogen loss Sources of nitrogen intake include meat, dairy, eggs, nuts and legumes, and grains and cereals. Examples of nitrogen losses include urine, feces, sweat, hair, and skin. Blood urea nitrogen can be used in estimating nitrogen balance, as can the urea concentration in urine. Nitrogen is a fundamental component of amino acids, which are the molecular building blocks of protein. Therefore, measuring nitrogen inputs and losses can be used to study protein metabolism. Positive nitrogen balance is associated with periods of growth, hypothyroidism, tissue repair, and pregnancy. This means that the intake of nitrogen into the body is greater than the loss of nitrogen from the body, so there is an increase in the total body pool of protein. Negative nitrogen balance is associated with burns, serious tissue injuries, fevers, hyperthyroidism, wasting diseases, and during periods of fasting. This means that the amount of nitrogen excreted from the body is greater than the amount of nitrogen ingested. A negative nitrogen balance can be used as part of a clinical evaluation of malnutrition. Nitrogen balance is the traditional method of determining dietary protein requirements. Determining dietary protein requirements using nitrogen balance requires that all nitrogen inputs and losses are carefully collected, to ensure that all nitrogen exchange is accounted for. In order to control nitrogen inputs and losses, nitrogen balance studies usually require participants to eat very specific diets (so total nitrogen intake is known) and stay in the study location for the duration of the study (to collect all nitrogen losses). Because of these conditions, it can be difficult to study the dietary protein requirements of certain populations using the nitrogen balance technique (e.g. children).

And all apparatus must be glass to withstand the acids. Chemically, nitric acid is made by bubbling nitrogen dioxide into water. So the objective in this approach is to generate nitrogen dioxide. This can be done by reacting hydrochloric acid, a nitrate salt and copper. Around 80grams of sodium nitrate, over 30 grams of copper and 100mL of hydrochloric acid (37% 12M) are the quantities needed. The exact amount isn't critical. For usable concentrations, the amount of water being converted should be small, around 20-50mL. Any source of nitrate is usable including potassium nitrate, ammonium nitrate and even nitrate-based fertilizers. Two approaches are shown in the video. In the first approach three containers, such as jars are place inside each other to force the gas to go into the water. This is very inefficient but is very simple to do. The better approach is to lead the gas out of the generator through a tube and into a chilled container of water. The water that's converted into nitric acid can be replaced with hydrogen peroxide for better yield. The chemical waste that's generated contains the valuable copper used before and recovering it is worthwhile due to todays high copper prices. This is simply done by putting in enough aluminum metal that it reacts with all the acids and copper in solution to create a slurry of copper. This can be filtered to obtain a residue of copper. Its highly contaminated but can still be used to make more nitric acid. For further information on the chemistry type "copper and nitric acid" into google. The hydrochloric acid and nitrate salt behave as nitric acid (with nitrate from the salt and protons from the hydrochloric acid) and dissolve the copper releasing nitrogen dioxide gas. You can use other concentrations of hydrochloric acid but you need to decrease the amount of water added to keep the concentrations the same. Finally, the last way of making pure nitric acid is to react concentrated sulfuric acid and a pure nitrate salt (NOT fertilizer) and heat it in a glass distillation apparatus to distill over the pure nitric acid. Red blood cells under the microscope, hypo and hypertonic solutions

Red blood cells (RBCs) as seen under the microscope in isotonic, hypotonic and hypertonic solutions. A few white blood cells can also be seen with the red blood cells. RBCs contain hemoglobin, a red pigment that binds to oxygen and allows oxygen to travel around the body. When RBCs are placed in a hypertonic salt solution, water moves from inside the cell to the outside by osmosis. This causes the cells to shrink. When RBCs are placed in distilled water, a hypotonic solution, water moves from outside of the cell to the inside, causing the cell to swell and rupture. Once the RBC ruptures, it loses its hemoglobin and becomes transparent and nearly invisible. These ruptured cells are called RBC ghosts and are very difficult to see in the video. I use two microscopes recorded through a Nikon D3300 DSLR camera.
